Abstract: The present study was carried out to develop a tranexamic acid-loaded liposomal gel for topical treatment of melasma. Tranexamic acid-loaded liposomes were prepared by the thin-film hydration method using different drug-to-lipid ratios, cholesterol and Tween 80 concentrations. The prepared liposomes were evaluated for vesicle size, PDI, zeta potential, entrapment efficiency and drug content. L5 was selected as the optimized formulation. The optimized liposomes were incorporated into Carbopol 940 gel. The final gel showed good appearance, homogeneity, pH, viscosity, spreadability and drug content. The drug content was 96.84 ± 0.78%. In-vitro drug release after 12 hours was 84.26 ± 1.72%, showing slower release than the plain gel. The developed tranexamic acid liposomal gel showed satisfactory properties and may be useful for topical delivery in melasma.
